Superfast drugs target shape-shifting enzymes

It all happens in a femtosecond ? a quadrillionth of a second. That's the time an enzyme needs to shape-shift into its most reactive form, trigger a chemical reaction and snap back into its original shape. We can now enter this high-speed world to interrupt the chemical reactions that sustain some of our deadliest pathogens and cause disease. Doing so could lead to antibiotics that won't trigger bacterial resistance.

In 1946, Nobel laureate Linus Pauling suggested that enzymes are most active during fleeting transition states, but only recently has Vern Schramm at the Albert Einstein College of Medicine in Yeshiva University, New York, moved the science from the blackboard to the clinic. "The problem in this whole field has been no one really knew what the structure of an enzyme's transition state looked like," he says. "It's hard to see something that has [effectively] no lifetime."

Over the course of a decade, Schramm has reconstructed those states, using computational and molecular modelling techniques. He has used the results to build drugs that bind so tenaciously to different enzymes' reactive forms ? like a baseball clamped to a catcher's mitt ? that the enzyme is essentially taken out of action.

One of his drugs neutralises a key enzyme in the malaria parasite, Plasmodium falciparum. Owl monkeys infected with usually lethal malaria cleared the illness after a week-long course of the drug.

Some of Schramm's drugs are at a more advanced stage. Two are in clinical trials ? one to treat gout and the other for leukaemia.

Lower dosage

Andrew Murkin of the University at Buffalo, New York, who formerly worked in Schramm's lab, is in the early stages of developing a similar drug, designed to tackle tuberculosis. He says that these drugs are so well tailored to block a specific enzyme that there is the potential to lower the dosage needed for efficacy. "You can't get that from other drug development approaches unless you're just plain fortuitous," he says.

Schramm's work may even hold the key to developing antibiotics that don't trigger resistance in bacteria. Traditional antibiotics lay waste to most bacteria, but some cells inevitably survive, and their mutated genes ? which are the source of resistance ? spread through the population.

To prevent this, we need antibiotics that cure the disease without killing the bacteria, so the bugs are not put under evolutionary pressure to mutate. Schramm has identified a promising target: an enzyme used in bacterial communication. We already know that bacteria which cannot communicate are far less virulent, but no less successful at eking out a living, than their communicating brethren.

Schramm has developed a drug that blocks an enzyme that Escherichia coli ? and the cholera-causing pathogen Vibrio cholerae ? use to communicate. Lab results are impressive: the drug is as effective against the 26th generation as it is against the first. The next step is to find a company to develop the drug further.

AP source: US brings new trade case against China

WASHINGTON (AP) ? The Obama administration is bringing a new trade case against China that seeks to pressure the rising economic power to end its export restrictions on key materials used to manufacture hybrid car batteries, flat-screen televisions and other high-tech goods.

The new trade initiative being announced Tuesday is another effort aimed at leveling the playing field for U.S. companies.

Senior Obama administration officials said the U.S. will ask the World Trade Organization to facilitate talks with China over its curtailment of exports of rare earth minerals. The U.S. is bringing the case to the WTO along with the European Union and Japan.

Tuesday morning, the EU announced its participation in the case. In Brussels, EU Trade Commissioner Karel De Gucht said China's restrictions "hurt our producers and consumers in the EU and across the world."

Obama was scheduled to speak about the WTO action from the White House later Tuesday, according to the U.S. officials, who requested anonymity in order to speak ahead of the president.

The fresh action is part of President Barack Obama's broader effort to crack down on what his administration sees as unfair trading practices by China that have put American companies at a competitive disadvantage.

China has a stranglehold on the global supply of 17 rare earth minerals that are essential for making high-tech goods, including hybrid cars, weapons, flat-screen TVs, mobile phones, mercury-vapor lights, smartphones and camera lenses. The materials also are used in the manufacture of tiny motors, such as those used to raise and lower car windows and in consumer electronics.

China has reduced its export quotas of these rare earth minerals over the past several years to cope with growing demand at home, though Chinese officials also cite environmental concerns as the reason for the restrictions. U.S. industry officials suggest it is an unfair trade practice, against rules established by the WTO, a group that includes China as a member.

The senior administration officials said Beijing's export restrictions give Chinese companies a competitive advantage by providing them access to more of these rare materials at a cheaper price, while forcing U.S. companies to manage with a smaller, more costly supply.

On Tuesday, a Chinese foreign ministry spokesman defended Beijing's curbs on rare earth production as necessary to limit environmental damage and conserve scarce resources.

"We think the policy is in line with WTO rules," said the spokesman, Liu Weimin, at a regular briefing.

He rejected complaints that China is limiting exports. "Exports have been stable. China will continue to export, and will manage rare earths based on WTO rules," Liu said.

The spokesman noted that China has about 35 percent of rare earth deposits but accounts for more than 90 percent of global production. "China hopes other countries can shoulder responsibility for supplies and can find alternative resources," he said.

Rare earth minerals are scattered throughout the Earth's crust, but only in small quantities, making them hard to mine. However, rich deposits of these rare earth oxides are in China, giving it command of the market.

The U.S. has just one rare earth mining company, the Colorado-based Molycorp Inc. There are also working mines in Australia, and a proposed one in Malaysia.

With the U.S economy slowly inching its way out of recession, Obama has sought to bring a renewed focus to Chinese policies that could hinder U.S. growth.

Obama used an executive order last month to create a new trade enforcement agency ? the Interagency Trade Enforcement Center ? to move aggressively against China and other nations. In announcing the new agency, Obama said it would bring "the full resources of the federal government to bear" in order to level the playing field for U.S. workers.

Under the terms of the WTO complaint, China has 10 days to respond and must hold talks with the U.S., E.U. and Japan within 60 days. If an agreement cannot be reached within that time frame, the U.S. and its partners could request a formal WTO panel to investigate Chinese practices.

The WTO, the only global international organization dealing with the rules of trade between nations, has sided with the U.S. in previous trade disputes with China.

In 2009 the Obama administration imposed a three-year tariff, starting at 35 percent, on U.S. imports of low-grade Chinese tires. The tariff was approved after imports of those tires rose threefold to about 46 million tires between 2004 and 2008. Last year the WTO rejected an appeal from China and found that the United States acted consistently with its obligations in imposing the duties.

Rambus Suffers Another Legal Blow

Technology licensing company Rambus Inc. (NASDAQ:RMBS) received an untoward ruling for a lawsuit filed for patent infringement, by a judge at the International Trade Commission (ITC). In the initial verdict, the judge mentioned that the plaintiffs did not conduct any breach with Rambus? patented technologies. However, the final judgment is due in July.

In a suit filed in 2010, Rambus complained that a number of its patents have been illegally used by chip makers LSI Corp. (NYSE:LSI), MediaTek Inc. and STMicroelectronics (NYSE:STM).

This is the second loss in a row for Rambus, which met a similar fate in November 2011. At that time, Rambus received an unfavorable ruling by a California jury on the long-drawn (almost 7 years) antitrust battle with Micron Technology Inc. (NYSE:MU) and Hynix Semiconductor Inc.

Micron and Hynix were accused of anti-trust charges, including conspiracy to fix DRAM prices and prevent Rambus? RDRAM memory technology from gaining traction in the market. However, considering the case a few months post trial, the jury negated the claims and cleared the defendants of all liabilities.

The unfavorable court decisions are jeopardizing investor sentiment, which is well inferred from the 3.5% decrease in share prices last Friday.

But, the good thing is that Rambus has been prompt in settling earlier claims with some tech companies. The company already made out-of-court settlements with NVIDIA Corp. (NASDAQ:NVDA), Freescale Semiconductor and Broadcom Corp. (NASDAQ:BRCM), signing patent licensing agreements with them. This arrangement not only saved some money for Rambus that might have been wasted in legal battle, but also secured its revenue streams.

Though Rambus? growth prospects are imminent, we remain on the sidelines given the Fed?s decision to declare three of its patents invalid. This would reduce its growth to the extent that it is dependent on the three patents. Moreover, Japanese chip-maker Elpida?s declaration of bankruptcy is also a concern for Rambus, as the company accounted for around 10.0% of its annual revenues. This would be a direct hit to its patent licensing royalty.

Currently, Rambus has a Zacks #3 Rank, indicating a short-term Hold rating.

Spider silk conducts heat as well as metals

ScienceDaily (Mar. 5, 2012) ? Xinwei Wang had a hunch that spider webs were worth a much closer look.

So he ordered eight spiders -- Nephila clavipes, golden silk orbweavers -- and put them to work eating crickets and spinning webs in the cages he set up in an Iowa State University greenhouse.

Wang, an associate professor of mechanical engineering at Iowa State, studies thermal conductivity, the ability of materials to conduct heat. He's been looking for organic materials that can effectively transfer heat. It's something diamonds, copper and aluminum are very good at; most materials from living things aren't very good at all.

But spider silk has some interesting properties: it's very strong, very stretchy, only 4 microns thick (human hair is about 60 microns) and, according to some speculation, could be a good conductor of heat. But nobody had actually tested spider silk for its thermal conductivity.

And so Wang, with partial support from the Army Research Office and the National Science Foundation, decided to try some lab experiments. Xiaopeng Huang, a post-doctoral research associate in mechanical engineering; and Guoqing Liu, a doctoral student in mechanical engineering, helped with the project.

"I think we tried the right material," Wang said of the results.

What Wang and his research team found was that spider silks -- particularly the draglines that anchor webs in place -- conduct heat better than most materials, including very good conductors such as silicon, aluminum and pure iron. Spider silk also conducts heat 1,000 times better than woven silkworm silk and 800 times better than other organic tissues.

A paper about the discovery -- "New Secrets of Spider Silk: Exceptionally High Thermal Conductivity and its Abnormal Change under Stretching" -- has just been published online by the journal Advanced Materials.

"Our discoveries will revolutionize the conventional thought on the low thermal conductivity of biological materials," Wang wrote in the paper.

The paper reports that using laboratory techniques developed by Wang -- "this takes time and patience" -- spider silk conducts heat at the rate of 416 watts per meter Kelvin. Copper measures 401. And skin tissues measure .6.

"This is very surprising because spider silk is organic material," Wang said. "For organic material, this is the highest ever. There are only a few materials higher -- silver and diamond."

Even more surprising, he said, is when spider silk is stretched, thermal conductivity also goes up. Wang said stretching spider silk to its 20 percent limit also increases conductivity by 20 percent. Most materials lose thermal conductivity when they're stretched.

That discovery "opens a door for soft materials to be another option for thermal conductivity tuning," Wang wrote in the paper.

And that could lead to spider silk helping to create flexible, heat-dissipating parts for electronics, better clothes for hot weather, bandages that don't trap heat and many other everyday applications.

What is it about spider silk that gives it these unusual heat-carrying properties?

Wang said it's all about the defect-free molecular structure of spider silk, including proteins that contain nanocrystals and the spring-shaped structures connecting the proteins. He said more research needs to be done to fully understand spider silk's heat-conducting abilities.

Wang is also wondering if spider silk can be modified in ways that enhance its thermal conductivity. He said the researchers' preliminary results are very promising.

And then Wang marveled at what he's learning about spider webs, everything from spider care to web unraveling techniques to the different silks within a single web. All that has one colleague calling him Iowa State's Spiderman.

"I've been doing thermal transport for many years," Wang said. "This is the most exciting thing, what I'm doing right now."

Even with Washington vote, focus remains on Ohio

(AP) ? Washington state was Saturday's prize for the Republican presidential candidates, but they focused on delegate-rich Ohio, among the 10 states holding contests on Super Tuesday in what will be campaign's biggest payday.

All four contenders have visited Washington ahead of the caucuses, but three of the four are campaigning in Ohio, the critical primary state next week. Mitt Romney, Rick Santorum and Newt Gingrich are fighting for the Midwestern state ? and it promises to be another important test for Romney, who seeks to quell doubts?about his candidacy from voters and the party establishment.

Santorum downplayed his expectations for Washington state while speaking Saturday morning at a rally that drew hundreds in Blue Ash, Ohio, a conservative corner of the state on the Kentucky border.

"We're trailing in the polls out there, but we feel like we might do pretty well," he said. "It would be a nice surprise heading into Super Tuesday's events here to have a good lift out of the state of Washington."

The former Pennsylvania senator repeatedly lashed out at Romney, charging the health care policy implemented while Romney was Massachusetts' governor had inspired President Barack Obama's health care bill and forced Catholic hospitals in the state to distribute the "morning-after pill."

"He is uniquely unqualified to go against Barack Obama in the biggest issue in this election," Santorum said.

Romney had criticized his rivals the night before at a rally in Cleveland.

"Let me tell you, the other guys, they spent their lives in Washington, working in a world of influence and in some cases lobbying," Romney said. "Except if you want to get the economy fixed and you want to create jobs, I?think it helps to have had a job. And I?have."

The Super Tuesday primaries and caucuses stretch from Vermont to Alaska ? where Texas Rep. Ron Paul was set to campaign Saturday ??but the top showdown is?in this?industrial state, a rematch of sorts after Michigan's primary. Romney won narrowly there over Santorum.

Romney flew to Ohio on Friday afternoon after a stop in Bellevue, Wash., where he held a campaign fundraiser and appeared at a rally at a community center.

"There are going to be a bunch of states that are going to make their mind up in the next couple days and so you guys are first," Romney?told an overflow crowd of hundreds. "Show up ? it won't take a long time, it'll just make a big difference."

Santorum, who visited Washington in February, was back Thursday for rallies in the more conservative eastern region, while Romney, who has been working to build support from establishment Republicans here and has rolled out dozens of local endorsements, hosted a high-dollar fundraiser in Bellevue, where tech giant Microsoft is based.

Their visits came on the heels of one by Gingrich last week.

Washington's caucuses are the last before Super Tuesday contests in Idaho, Alaska, Georgia, Massachusetts, North Dakota, Ohio, Oklahoma, Tennessee, Vermont and Virginia, offering a total of 419 delegates.

Washington's caucuses are an opportunity for Paul. He's?the only candidate on the air, having spent roughly $40,000 to run ads on cable channels.

At stake are 40 delegates to the Republican national nominating convention this summer, a cache second only to Florida's 50 in contests thus far. Registered voters of all political stripes can participate in the caucuses, but they must sign an affidavit identifying themselves as Republican and promising not to participate in a caucus for another party.

In Ohio, Romney, Santorum and Gingrich plan to participate in a forum hosted by Mike Huckabee and taping Saturday afternoon. Romney also?planned events in Dayton and Cincinnati. Santorum and Gingrich were both set to speak at a Lincoln Day dinner in Bowling Green and planned other events across the state.

SPIN METER: Santorum's shifting views on education

(AP) ? Republican presidential candidate Rick Santorum is scornful of the government's hand in public education, saying schooling is the task of parents and pointing out that he and his wife have home-schooled their seven children.

Yet back when Santorum was a senator from Pennsylvania, he got a Pittsburgh-area school district to help pay tens of thousands of dollars in tuition for his children to receive online schooling.

It's a bit of history that's unknown to most of those now hearing Santorum pitch for conservative votes he needs to overtake GOP frontrunner Mitt Romney.

Santorum says he wants to dramatically curtail the role states and the federal government play in running schools.

"Not only do I believe the federal government should get out of the education business, I think the state government should start to get out of the education business and put it back with the local and into the community," Santorum said in a recent debate in Arizona with his GOP rivals.

He mocks America's schools as "factories" that stand as "anachronistic" relics of the Industrial Revolution and says he would home-school his kids in the White House if he becomes president. The Los Angeles Times dubbed him perhaps "the most prominent" home-schooler in America.

In the fall of 2004, Santorum's use of tax dollars to pay for his kids' home schooling became controversial because his family was primarily living in Leesburg, Va., an outer suburb of Washington. Following a local newspaper report, the Penn Hills School District near Pittsburgh tried to recover about $73,000 that it contended the state wrongly sent to an Internet-based charter school because although the Santorums owned a house in the school district, they were living out of state. The Pennsylvania Education Department in 2006 agreed to pay the district $55,000 to settle the dispute.

The cyberschool controversy dogged Santorum through his 2006 Senate re-election bid and contributed to his 18-point loss to Democrat Bob Casey. Santorum's campaign did not respond this week to questions about his family's online instruction, and it's not known whether his children received teaching at home in addition to what they got online.

The Santorums withdrew their children from the cyberschool and resumed home schooling after Penn Hills officials complained about the tuition payments. Students in cyberschools log onto computers to access their assignments and teachers.

The National Home Education Research Institute, which specializes in home-school research, estimated in spring 2010 there were more than 2 million home-schooled students, about 3 percent of the school-age population. Brian D. Ray, president of the institute, said while he didn't know what percentage of those students use online charter schools, he has watched it grow significantly over the past five years.

Pennsylvania law requires school districts to pay for resident students who enroll in cyberschools, and Santorum at the time of the controversy said the Penn Hills house was his family's legal residence and that he paid taxes for it.

Erin Vecchio, a former Penn Hills school board member and former head of the local Democratic committee, at the time questioned whether it was proper for the school district to pay the cyberschool tuition for five of Santorum's children because they spent most of their time at his home in Virginia. She said Santorum should have reimbursed the district for the tuition costs.

"He should have been held accountable for that money, but he wasn't," Vecchio said in a recent telephone interview with the AP. "When he found a program that he could use to his advantage, he used it. That's the thing with Rick Santorum."

Vecchio said the fight over Santorum's residency was ironic, given how Santorum had made challenging the residency of Democrat Doug Walgren a key campaign issue when he toppled the incumbent and won his House seat in 1990. Santorum slammed the seven-term representative for living with his family in McLean, Va.

Santorum and his wife, Karen, now own a home in Great Falls, Va., an affluent Washington suburb. They moved there after Santorum's 2006 Senate loss.

Acknowledging that its own rules were confusing, the Pennsylvania Education Department in 2006 agreed to settle the dispute by repaying the district. The state Education Department said the money was not a reimbursement, but an acknowledgment that the department gave conflicting rules about when a district can challenge the state's decision to withhold cyberschool tuition fees from the district.

On the campaign trail, Santorum's candidacy has been boosted by Christian home-school advocates, evangelical pastors and tightly knit networks of conservative activists who helped him win Iowa's leadoff caucuses and a three-state sweep of contests on Feb. 7. Limited government has been a big part of his pitch.

Santorum now says he regrets voting for the sweeping No Child Left Behind education overhaul. He's called for a significantly smaller Education Department but would not eliminate it. He's also criticized early childhood education programs as an attempt by government to "indoctrinate your children."

Santorum says government can only do so much to educate kids, and that parents bear the prime responsibility.

"Yes, the government can help," Santorum said during a recent stop in Ohio, which holds a key contest in the upcoming March 6 Super Tuesday contests. "But the idea that the federal government should be running schools, frankly much less that the state government should be running schools, is anachronistic."

He said it dates from the nation's industrialization, "when people came off the farms where they did home-school or have the little neighborhood school, and into these big factories, so we built equal factories called public schools. And while those factories as we all know in Ohio and Pennsylvania have fundamentally changed, the factory school has not."
